Next generation Mendelian genetics in familial Alzheimer disease Lead Investigator: Zoran Brkanac Institution : University of Washington E-Mail : zbrkanac@uw.edu Proposal ID : 385 Proposal Description: The central hypothesis for this proposal is that Mendelian causes of familial Alzheimer disease (FAD) in existing samples can be identified using novel analytic approaches and next generation sequencing technologies. We propose to use exome sequencing in NCRAD families with 4 or more cases where the mode of transmission of Alzheimer disease (AD) is consistent with a single gene effects. We expect that for each analyzed family we will identify only a handful of candidate genes that are likely to be causal. Next, we will use a ???mutational load??? case-control study with NCRAD and other Alzheimer familial cases to establish the association of identified genes with AD. This will be accomplished through following specific aims. Aim 1: Exome sequencing and analysis. Aim 2: Sequence candidate genes for association with Alzheimer disease.
